Analyze phosphoinositides in tissue and cultured cells

I have used Thermo and AB Sciex triple quadropole instruments in addition to Waters Xevo to measure cellular phosphoinositides that have been derivatized with trimethylsilyl-diazomethane. The Waters Xevo TQ S with Acquity UPLC and MassLynx software tops them all for ease of use and reproducibility. While they may all have adequate sensitivities, the Xevo delivers the most consistent results with the least amount of fuss. MassLynx, while not without its glitches is by far the most intuitive and easy to use when compared with AB Sciex and Thermo Xcalibur software. Although, I was not the purchaser, this is the brand I would purchase if given the choice.

A comparison of Extraction Techniques in Bioanalysis and Forensic Toxicology Analyses

In this application note, a comparison was performed between Oasis PRiME HLB SPE, SLE and LLE in both plasma and urine matrices for bioanalysis and forensic toxicology. In plasma, 22 comonly analyzed pharmaceuticals, steroids, and drugs of abuse were extracted using the three aforementioned methods and the results were compared. In urine, 23 drugs of abuse representing opioids, stimulants, benzodiazepines, and synthetic cannabinoid metabolites were tested for forensic toxicology analysis.

Analysis of Ketamine and Xylazine in Rat Tissues

In Veterinary medicine, xylazine is an FDA approved tranquilizer often used in combination with ketamine. The recreational use of both drugs is on the rise in a number of countries. Recently, the cocktail has been reported as a date rape drug resulting in ketamine being re-classified as a Schedule III drug in the US. the analysis of xylazine and ketamine in biological samples presents a number of challenges to analysts. Hyphenated chromatography platforms have enabled the sensitivity and efficiency to detect trace levels of analytes previously unachievable by traditional means. The only bottleneck now is the lengthy sample preparation time required, which will inevitably delay forensic investigations.

Highly Sensitive Analysis of Polar Pesticides in Food Matrices on the Xevo TQ-XS

This application note describes a sensitive LC-MS/MS quantification method for the robust analysis of highly polar pesticides in food commodities without the need of derivatization. Using the ACQUITY UPLC System coupled to the Xevo TQ-XS Triple Quadrupole Mass Spectrometer, they achieved sub-parts per billion detection (ppb) limits for glyphosate, glufosinate and aminomethylphosphonic acid (AMPA). The sensitivity of the Xevo TQ-XS promises excellent limits of detection, suitable for monitoring maximum residue limit (MRL) compliance for these challenging analytes and good quantitative performance, in terms of precision and calibration characteristics in compliance with SANTE 11954/2015 guidelines.
