Selective Extraction and Low Level Quantification of the Mycotoxin Patulin from Organic Apple Puree

Selective Extraction and Low Level Quantification of the Mycotoxin Patulin from Organic Apple Puree

19 Nov 2015

Patulin is a mycotoxin that commonly infects apples used to produce juice and other apple products. Exposure to patulin may negatively impact human health so it is regulated in many countries including the United States and European Union member states. A method utilizing SupelMIP® SPE- Patulin cartridges and a Titan™ C18 UHPLC column with UV detection was developed to successfully demonstrate the extraction, detection, and quantification of patulin at low concentrations in organic apple puree.

SupelMIP® SPE - Patulin

Sigma-Aldrich Supelco

The SupelMIP SPE line consist s of highly cross-linked polymers that are engineered to extract a single analyte of interest or a class of structurally related analytes with an extremely high degree of selectivity. This is possible because selectivity is introduced during MIP synthesis in which a template molecule, designed to mimic the analyte, guides the formation of specific cavities or imprints that are sterically and chemically complementary to the analyte(s) of interest.By careful design of the imprinting site, either by molecular modeling, experimental design, or screening methods, the binding cavities can be engineered to offer multiple interaction points with the analyte(s) of interest. This leads to a stronger interaction between the solid-phase and the analyte(s). As a consequence, harsher wash conditions can be tolerated during SPE methodology resulting in cleaner extracts. Because extraction selectivity is significantly improved, lower background is observed allowing analysts to achieve lower limits of detection.Features & Benefits: Highly selective SPE for trace analysis in complex matrices Achieve lower detection limits Significant time and cost savings Improved MS-compatibility

Titan™ C18 UHPLC Columns, 1.9 μm

Sigma-Aldrich Supelco

Titan UHPLC columns, based on 1.9 µm totally porous monodisperse silica, outperform other UHPLC columns. These UHPLC columns provide the performance scientists expect (typically > 250,000 N/m), but at a greatly reduced cost. With the narrowest particle size distribution of all available sub-2 µm porous particles, Titan columns are the outcome of the highly efficient patent pending Ecoporous™ silica manufacturing process that delivers high-performance silica at a low cost. Read about how this product was used to analyze Riboflavin and impurities here and used in purification methods here. Features & Benefits: • Excellent Performance • Extremely Narrow Particle Size Distribution • Great Value

MicrobiologyMicrobiology is the study of microorganisms including protists, prokaryotes, fungi, and, often, viruses. Microorganisms are a useful research tool as genetic vectors and, in immunology, for antibiotic susceptibility testing, cellular biology and genetics. Microorganisms commonly grow readily in incubators with microbial culture media; this can contain chromogenic supplements to differentiate between cell lines. Estimate your culture’s density of microorganisms with colony counters, or screen and select colonies for desirable clones with automated colony pickers. Additionally, equipment is available to monitor environments for the presence of microbes and identify with microbial identification instruments. Multiple formats are available for conducting SPE, including prepacked SPE cartridges, disks and microplates, as well as SPE sorbent powders for manual packing. SPE systems can be used to automate the process and extract multiple samples at once. Solid-phase microextraction (SPME) and supported liquid extraction (SLE) products are also available in the same formats.
