NPN and NCN determination in milk according to the Dumas combustion method
23 May 2021Milk is a source of animal protein of high nutritional quality. The nitrogen compounds of milk can be divided in: casein proteins and non-casein nitrogen (NCN). Among this, there are soluble whey proteins (WP), enzymes and non-proteins nitrogen substances (NPN), composed of urea, amino acids, uric acid, creatine, creatinine and ammonia. Although Kjeldahl is nowadays the most common method for determining nitrogen and protein contents in foods and feeds, Dumas method, shows a high level of precision and reproducibility compared to Kjeldahl method.

KjeldahlKjeldahl is an analytical chemistry technique for the quantitation of nitrogen in chemical substances. Applications of Kjeldahl include analyzing the protein content of food, nitrogen analysis in environmental sciences and the pharmaceutical industry and total kjeldahl nitrogen, nitrate and ammonia analyses in agricultural sciences.Food and Beverage ProcessingFood and Beverage Processing involves the preparation of food products for further analysis. Typical instrumentation includes blanchers, color sorters, cookers, grinders, and refrigeration. These are used to replicate the processes commonly used in the commercial preparation of food and beverages.Protein QuantificationThe detection and quantification of proteins in a sample is vital across life sciences, pharmaceutical research and clinical diagnostics, and a variety of equipment is available to scientists to simplify the workflow. Proteins of interest can be easily labeled and detected on light-based detection instruments. Immunoassay kits allow you to identify a specific protein of interest and protein detection beads or antibody microarrays allow you to identify multiple specific proteins at once. Protein interactions and enzyme activity can also be monitored with protein-protein interaction assays. Additionally, the biophysical characterization of proteins is made easy with biokinetic analyzers. It is consumed by infant mammals to provide nutrients from the mother. In 2008, the Chinese milk scandal saw the unlawful adulteration of powdered milk with melamine.Food TestingFood testing refers to a variety experiments including PCR, mass spectrometry, processing, QuEChERS, analyzing and Kjeldahl.
