Introducing the RNAscope Plus small RNA-RNA fluorescent assay

23 Apr 2023

In this application note, Advanced Cell Diagnostics introduces the RNAscope™ Plus smRNA-RNA fluorescent assay, a sensitive technology for detecting small regulatory RNA alongside target RNA in intact cells/tissues. This assay can visualize one smRNA target and three RNA targets simultaneously and employs four independent signal amplification systems using different fluorophores. It has a two-week turnaround time for custom designs with over 42,000 pre-designed probes available. The assay has demonstrated detection of positive control miRNA and RNA targets in HeLa cells and mouse colon FFPE tissue, tissue-specific detection of miR-124 in mouse brain tissue, and colocalization of miRNA and associated mRNA targets in head and neck cancer tissue.

Fluorescence SpectroscopyFluorometers and spectrofluorometers (also called fluorescence spectrometers) are used to measure the intensity and wavelength of fluorescent light emitted from a sample after excitation by illumination. Spectrofluorometers utilize monochromators to select the desired wavelengths, whereas filter fluorometers employ a set of filters. Spectrofluorometers for measuring steady-state fluorescence and lifetime fluorescence (or time-resolved fluorescence) are available, as well as fluorescence microscopes and microplate readers. This can reveal information on cell health (apoptosis, cytotoxicity, viability and proliferation assays), cell metabolism, cell migration and cell signaling mechanisms. Fluorescence can be detected and measured for different purposes using microplate readers, fluorescence microscopes, fluorescence scanners, and flow cytometers.Fluorescence Based AssayFluorescence based assays are widely used in life science research and high-throughput screening to measure a broad range of cellular activities.RNARNA is a nucleic acid that plays a key role in gene expression and protein synthesis. It serves as a messenger between DNA and ribosomes, carrying genetic instructions to produce proteins. Advances in RNA-based therapies, such as mRNA vaccines and gene editing, have revolutionized treatment strategies for genetic disorders and infectious diseases.
