Detection and Cross Contamination Evaluation of Roundup Ready Soy via Mechanical Shear Homogenization

5 Oct 2017

Genetically modified crop are food products that have had their DNA modified to introduce new traits such as resistance to pests, diseases or environmental conditions. In this application note, Round-up Ready® soy beans are homogenized so DNA can be purified and the Roundup resistant gene can be detected. This process is carried out using the GLH-850 by Omni International in order to evaluate its homogenization capabilities.

GLH 850 General Laboratory Homogenizer

OMNI International Inc.

The GLH 850 is an affordable, high performance instrument capable of processing virtually any laboratory sample in seconds. A powerful 850 watt motor provides maximum versatility for homogenizing, dispersing or emulsifying the largest variety of sample types and sizes in its class. The GLH 850 features an advanced, integrated, push button digital control system that ensures accuracy and repeatability. The unit’s closed loop variable speed controller (5,000 - 25,000 rpm) maintains selected speeds within +/- 100 rpm of the original set point. The digital timer can be set from seconds to minutes, and includes a 30 second countdown timer, which is ideal for remote applications in fume hoods or biosafety cabinets. A soft start feature ensures smooth processing and minimizes splashing. The GLH 850 is compatible with a wide range of interchangeable stainless steel rotor stator generator probes (sold separately), from 5 mm to 30 mm diameter, and processes samples ranging from Patented Omni TipTM Plastic Generator Probes, in both soft tissue and hard tissue versions, are available for researchers concerned about cross contamination between samples. Stainless steel generator probes are recommended for larger, tougher samples. Specially designed Hybrid Plastic/Stainless steel probes are also available.

DNA / RNA Extraction and PurificationPurified DNA and RNA are required for numerous downstream molecular biology applications. Consequently, the importance of high-quality DNA/RNA extraction and purification equipment cannot be underestimated. Many purification kits are available and are typically optimized for nucleic acid type and source, including plasmid DNA, genomic DNA, mRNA, RNA and viral nucleic acid purification kits. Automated extraction and purification of nucleic acids can be implemented with magnetic bead separator instruments or high-throughput purification workstations. Lab equipment for sample homogenization includes handheld and benchtop homogenizers, bead mill, ultrasonic and multiple sample homogenizers, high shear homogenizers and blenders. Homogenizer bead and probe multipacks are available to avoid cross contamination of samples, as well as cooling systems for homogenizing temperature-sensitive samples. Homogenizers are often used in the first step of extraction of DNA, RNA and proteins from tissue and cell samples.
