This application note describes the configuration and preprogrammed methods for the automated construction of 8/16 or 24 sequencing ready libraries from 100-1000ng total RNA using the Illumina TruSeq Stranded Total RNA kit. The overall hands on time is less than 1 hour, the total run time of the entire procedure is ~11.5 hours for 24 samples.
