Assessing Aggregation and Kinetics of Liposome-Protein Complexes using NanoSight Technology

25 Oct 2016

This whitepaper describes rapid characterization of liposome-insulin receptor complexes by Nanoparticle Tracking Analysis using the NanoSight NS300 from Malvern Instruments. The NanoSight technology observes the Brownian motion of particles in suspension and calculates their size through the Stokes-Einstein equation. The paper concludes that the high resolution NTA using the NanoSight NS300 is a useful technique in analyzing the aggregation kinetics of liposome preparations.

NanotechnologyNanotechnology, or nanotech, is an engineering technique using molecular scale functional systems. Applications of nanotechnology include medicine and medical devices, electronics, air and water purification, food science and energy production.Particle CharacterizationParticle characterization instruments are used to determine particle size distribution, shape, surface area, zeta potential, density and porosity of particles and materials. Multiple tecchniques are available for determining particle size, shape and count including dynamic light scattering (DLS), laser diffraction, electrozone (Coulter technique), imaging particle analysis and single particle optical sensing. Determine the density of your material with a gas pycnometer or examine its surface area and porosity with gas adsorption analyzers and mercury porosimeters. Drug delivery is often approached via the biopharmaceutical or small molecule drug's formulation, but it may also involve medical devices or drug-device combination products. Considerations include instrumentation, software and services. AggregationHigh ThroughputHigh throughput experiments allow the simultaneous processing of several samples. This parallelization reduces the cost per experiment and increases reproducibility and output volume of data.ReceptorsInsulinPharmaceuticalsPharmaceuticals are medicinal drugs used in healthcare to diagnose, prevent, cure and treat illnesses. Pharmaceuticals that are excreted after use appear in wastewater and can have detrimental effects on the environment.NanoparticlesNanoparticles are between 1-100nm in size. Nanoparticles can be used for a wide variety of applications including biomedical, catalysis and electronics.Drug DevelopmentDrug development refers to the process of bringing a new drug to market.
