Advanced cell analyses of mammalian cells using the NucleoCounter NC-3000'

12 Jul 2021

Cell death can occur by two distinct mechanisms, necrosis or apoptosis. Necrosis occurs when cells are exposed to harsh physical or chemical stress (e.g., hypothermia, hypoxia) while apoptosis is a tightly regulated biochemical process by which cells are eliminated and where the cell is an active participant in its own termination (“cellular suicide”). This application note demonstrates how to use NucleoCounter® NC-3000™ to detect cells with caspase activity using the Fluorochrome-labelled Inhibitor of Caspases Assay.

NucleoCounter® NC-3000™ advanced cell analyzer

ChemoMetec

The NucleoCounter® NC-3000™ is an advanced image cytometer utilizing fluorescence imaging to characterize cell properties. The NC-3000™ can perform high-speed cell count and viability analyses and it includes plug-and-play analytical assays of mammalian, yeast and insect cells. Pre-defined assays include a unique 5-minute cell cycle assay, a selection of apoptosis assays and a GFP transfection efficiency assay.

Flow Cytometry / Cell CountingFlow cytometers are used to count, sort and examine multiple characteristics of cells. Other cell analysis equipment includes image cytometers, cell counters, fluorescence-activated cell sorters (FACS), magnetic-activated cell sorters (MACS), and a range of flow cytometry assay kits. Flow cytometers can reveal information on cell viability, cell proliferation, apoptosis and cell cycle progression, as well as identify cell populations and intracellular or cell-surface molecules. Additionally, some flow cytometers, known as FACS, have an additional sorting function after analysis. Cell counters and image cytometers count live and dead cell populations and can also conduct cell proliferation assays. Sample preparation and processing includes fixation, staining, sectioning and slide mounting, using equipment such microtomes and cryostats. In choosing immunohistochemistry and immunocytochemistry kits, consider chromogens, staining method, antibodies, microscopes and imaging.ApoptosisApoptosis, or programmed cell death, is a vital process for healthy tissues and is key to our understanding of disease mechanisms like cancer and neurodegeneration. Research tools enable apoptosis detection, quantification, and analysis. Fluorescence can be detected and measured for different purposes using microplate readers, fluorescence microscopes, fluorescence scanners, and flow cytometers.CytometryCytometry is a broad term for the quantitative analysis of cells and cell systems. The most popular methods to study cytometry are flow cytometry and image cytometry.Cell AnalysisThe analysis of cells allows researchers to understand the factors which contribute to cell health and function. These influencing processes can then be predicted and altered, leading to the development of medication and disease treatments.
