Reliable with good i.c. staining properties
Flow Cytometry
Always stains the right population, with early thymocytes negative, and later thymocytes staining nicely. Works really nicely of intracellular flow, with the antibody staining icTCRb + cells at the expected developmental stage and little background. Shift between -ve and +ve can sometimes be small though, and fluorochrome does not stand up well to harsh conditions (e.g. methanol fix/perm protocols, prolonged periods at 37 deg. C).
