Histone H3 is one of the core components of the nucleosome, the basic building block of chromatin. Lysine 79 methylation in yeast is dependant upon the ubiquitylation of lysine 123 of histone H2A. H3 lysine 79 methylation has been implicated in transcriptional activation, transcriptional silencing and (through the recruitment of the 53BP1 protein) DNA repair.
