Yarra 1.8μm SEC-X150 Columns - Superior Stability under Harsh Conditions

21 Jul 2016

This application note demonstrates the performance advantages of Yarra 1.8μm SEC-X150 Columns from Phenomenex for the analyses of biomolecules by aqueous size exclusion chromatography (SEC). The switching of mobile phases was used to simulate stress on the silica bed. Faster flow rates served to increase the backpressure to atypical levels and the stoppage of flow after each series of steps mimicked the completion of a series of samples. Column failure was not actually achieved after this study was completed, modeling the stable performance of Yarra SEC-X150 columns over longer lifetimes.

Yarra Ultra-High Efficiency Aqueous GFC/SEC Columns

Starting with 3 µm ultra-pure silica, Yarra particles are densely bonded with a proprietary surface chemistry. Coupled with tight particle and pore size distribution as well as strict packing and QC specifications, Yarra GFC columns allow for the highest efficiencies and resolution possible. Yarra columns are offered in three phases (SEC-2000, 3000, and 4000), and are ideal for the separation of small to large proteins, as well as biological therapeutics and biosimilars.

UHPLC and HPLCHigh performance liquid chromatography (HPLC) and ultra high performance liquid chromatography (UHPLC), also known as UPLC, are analytical techniques used to separate, identify and quantitate components of complex mixtures including biological samples such as proteins and lipids as well as chemical mixtures of pesticides, drugs and oils. Both techniques are liquid chromatographic methods but differ by operating pressures (HPLC < 6000 psi < UHPLC ). Components of HPLC and UHPLC systems include columns, detectors, pumps, autosamplers and column heaters. Explore a range of UHPLC and HPLC columns for your specific sample needs including reverse phase, normal phase, ion exchange, HILIC, ion exclusion and size exclusion columns. SEC is further subdivided into gel filtration chromatography (GFC) and gel permeation chromatography (GPC) when an aqueous or organic mobile phase is used respectively. HPLC-based SEC systems and columns are available for analytical separation alongside SEC spin columns or filters to prep samples for many protocols. It is widely used in fields like biochemistry, pharmaceuticals, and environmental analysis. By exploiting differences in the interaction of substances with a stationary phase and a mobile phase, chromatography enables precise purification and quantification of compounds. Whether you're working with complex biological samples or industrial chemicals, chromatography is essential for obtaining high-quality results.
