Studying Paneth Cell Differentiation Using the Tecnai with iCorr

9 Apr 2015

Intestinal Paneth cells, originally identified as an anti-microbial secreting cell population, have emerged as critical modulators of active cycling intestinal stem cells, supplying critical molecular cues that influence stem cell behavior. In a study to elucidate factors that influence Paneth cell differentiation, Nick Smith and Melissa Wong (OHSU) used the Tecnai with iCorr to perform correlative light and electron microscopy imaging of mouse intestinal tissue to evaluate the full differentiation of the Paneth cell population.

Light MicroscopyLight microscopes or optical microscopes are used to visualize microscale objects under magnification, including cells, clinical specimens and materials. Lab equipment for light microscopy includes confocal microscopes, fluorescence microscopes, zoom and stereo microscopes. Microscope slides and imaging reagents are available for visualizing samples, as well as various microscope stages and incubators for large or temperature-sensitive samples. Types of electron microscope include scanning electron microscopes (SEM), transmission electron microscopes (TEM), scanning transmission electron microscopes (STEM) and cryo-electron microscopes. Focused ion beam (FIB) microscopes are useful for modifying or milling a sample surface with nanometer precision, as well as imaging.
