Simplified workflow using automated instrument setup and compensation on the BD FACSLyric Flow Cytometer

21 Feb 2024

In this application note, immunophenotyping results from three to six 10-color BD FACSLyric™ Flow Cytometers were evaluated over two months. Overall performance was assessed using a multicolor assay for T, B and NK cells in peripheral blood from HIV patients and controls. The results show that the BD FACSLyric™ System provides unique tools to the clinical user to help achieve reproducible and consistent results across time and instruments. The application note demonstrates the ease of use, reproducibility of automated setup and compensation, and resolution sensitivity of the BD FACSLyric™ Flow Cytometer platform. These features may be useful for generating consistent results for longitudinal or multisite studies in which the same assay is used to acquire data on different days, on different instruments or by many operators with varying levels of expertise.

Fluorescence SpectroscopyFluorometers and spectrofluorometers (also called fluorescence spectrometers) are used to measure the intensity and wavelength of fluorescent light emitted from a sample after excitation by illumination. Spectrofluorometers utilize monochromators to select the desired wavelengths, whereas filter fluorometers employ a set of filters. Spectrofluorometers for measuring steady-state fluorescence and lifetime fluorescence (or time-resolved fluorescence) are available, as well as fluorescence microscopes and microplate readers. Other cell analysis equipment includes image cytometers, cell counters, fluorescence-activated cell sorters (FACS), magnetic-activated cell sorters (MACS), and a range of flow cytometry assay kits. Flow cytometers can reveal information on cell viability, cell proliferation, apoptosis and cell cycle progression, as well as identify cell populations and intracellular or cell-surface molecules. Additionally, some flow cytometers, known as FACS, have an additional sorting function after analysis. Cell counters and image cytometers count live and dead cell populations and can also conduct cell proliferation assays. Software is used for analysis of ADME results, toxicology, clinical trials and regulatory processes. FluorescenceThe emission of fluorescence occurs when a photon of energy is supplied to a fluorescent chemical compound by an external source, causing it to become excited. Fluorescence can be detected and measured for different purposes using microplate readers, fluorescence microscopes, fluorescence scanners, and flow cytometers.Single Cell AnalysisSingle-cell analysis involves studying individual cells to gain insights into their behavior, gene expression, and function. This approach is valuable in cancer research, stem cell biology, and immunology.
