Practical guide on key considerations – Deprotection to completion

Glen Research has created an introductory guide on oligonucleotide synthesis for newcomers. This practical guide details the various considerations within the field of oligo synthesis, and focuses on three parts that can be visualized: Cleavage, phosphate deprotection, and base deprotection. Cleavage is removal from the support, phosphate deprotection is the removal of the cyanoethyl protecting groups from the phosphate backbone, and base deprotection is the removal of the protecting groups on the bases or modifier.

It is however not the intent of the guide to provide a comprehensive, fully referenced review of deprotection strategies in oligonucleotide synthesis – they are simply guidelines.
