Neural outgrowth analysis in 3D human brain micro-tissues for compound screening applications

24 Mar 2024

Organotypic, three-dimensional (3D) cultures, such as brain organoids, offer higher in vivo relevance compared to traditional 2D cultures for studying neurological development and disease. In this application note, Molecular Devices highlights RealBrain® micro-tissues by Tessara Therapeutics which provide greater physiological relevance and scalability for analyzing 3D neurite outgrowth. In a study using RealBrain micro-tissues, compounds affecting neurite outgrowth were imaged using high-content imaging and analyzed with MetaXpress® software.

Cell / Tissue CultureCell culture or tissue culture is used to study the biology of cells or tissues and to isolate cellular products in an environment which can be manipulated and well defined. Accurately control your culture environment with bioreactors or culture incubators, bind your cells to a surface or together with an extracellular matrix. Distinguish cell types with differential media or proliferate cells with certain characteristics using selective media. Enrich your media with supplements such as growth factors, sera and vitamins. Use HTS readers and integrated assay preparation / analysis workstations to screen your compounds. Identify active compounds against various HTS libraries, including membranes, proteins and peptides and HTS cell lines. HCS uses fluorescent microscopic imaging and automated image analysis to investigate cellular events such as apoptosis, cell viability, GPCR activation, oxide production, neurite outgrowth, and cell signaling. Find the best fluorescent labeling reagents, cellular assays, and high-content imaging systems in our peer-reviewed product directory: compare products, check customer reviews and receive pricing direct from manufacturers. In Vivo Imaging SystemsIn vivo imaging systems, including pre-clinical imaging systems and medical imaging systems are used to non-invasively visualize and capture images of live animals and plants. Monitor the natural processes or diseases of your subjects using small-animal pre-clinical imaging systems, including single photon positron emission tomography (SPECT), positron emission tomography (PET), computed tomography (micro-CT), magnetic resonance imaging (MRI), X-ray radiography, ultrasound, fluorescence and bioluminescence imagers. Multimodal systems and software solutions are also available for correlative analysis of organ, tissue, cell, or molecular-level processes. The process is adapted to multi-well plates and both the image acquisition and analysis are automated.Drug DiscoveryDrug discovery is the process of identifying potential new medications, involving stages such as target identification, compound screening, and preclinical development. It relies on cutting-edge technologies like high-throughput screening, artificial intelligence, and molecular modeling to accelerate the identification of drug candidates. Drug discovery plays a pivotal role in developing new therapies for diseases ranging from cancer to rare genetic disorders.
