InCELL Hunter: A Novel Cellular Assay Platform for the Direct Measurement of Compound Binding to Epigenetic and Kinase Targets in Intact Mammalian Cells
23 Sept 2012This poster, presented at ELRIG Drug Discovery 2012, describes how the InCell Hunter™ platform from DiscoveRx Corporation can be used to detect the binding of a small molecule to a protein target of interest in intact mammalian calls by monitoring changes in protein structure and stability. The system has been used to develop a large panel of cell-based kinase, bromodomain and methyl transferase assays for screening and profiling. The InCell Hunter™ platform has the potential to enable the development of cell-based assays for difficult kinase targets including low turnover enzymes and targets whose substrates have not yet been identified.

Assay AssemblyAssay Assembly is technique used in drug discovery to develop assays to test the cytotoxicity, genotoxicity, or other activities of a compound on a cell. Assay assembly requires chip assembly, a delivery system and a detection and analysis method. Beneficial features of assay kits or automated systems include high-throughput, high speed and sensitivity and low signal to noise ratio.Cell-Based AssaysCell-based assays are used to monitor the presence, quantity and activities of a desired cellular analyte including drug molecules or biomarkers. This can reveal information on cell health (apoptosis, cytotoxicity, viability and proliferation assays), cell metabolism, cell migration and cell signaling mechanisms.
