Homogeneous Add-Mix-Read Assay for Total and Oxidized Glutathione

27 Aug 2015

A wide variety of compounds can induce intracellular formation of reactive oxygen species (ROS) and the resulting cellular damage can eventually lead to apoptosis or necrosis. Measuring the ratio of reduced to oxidized glutathione (GSH/GSSG ratio) is a way to determine how such compounds alter the redox potential in the cell. GSH/GSSG-Glo allows for rapid, direct measurement of total and oxidized glutathione in cell-culture wells using low cell density.

GSH-Glo™ Glutathione Assay

Promega Corp.

The GSH-Glo™ Glutathione Assay is a luminescence-based assay for the detection and quantification of glutathione (GSH). The assay is based on the conversion of a luciferin derivative into luciferin in the presence of glutathione, catalyzed by glutathione S-transferase (GST). The signal generated in a coupled reaction with firefly luciferase is proportional to the amount of glutathione present in the sample. Luminescence is generated in less than one hour. The assay generates a stable luminescence signal and is simple, fast and easily adaptable to multiwell formats such as 96- and 384-well plates. This assay can be used for detection and quantification of GSH in cultured cells or in various biological samples.

Cell / Tissue CultureCell culture or tissue culture is used to study the biology of cells or tissues and to isolate cellular products in an environment which can be manipulated and well defined. Accurately control your culture environment with bioreactors or culture incubators, bind your cells to a surface or together with an extracellular matrix. Distinguish cell types with differential media or proliferate cells with certain characteristics using selective media. Enrich your media with supplements such as growth factors, sera and vitamins.
