High-Throughput, High-Resolution Monoclonal Antibody Analysis with Small Particle Size HPLC Columns

17 Feb 2015

This application note shows the selectivity differences between ProPac WCX-10 and MAbPac SCX-10 phases for protein separation. Applications using smaller particle columns for high-throughput, high-resolution mAb separations are also described. The ruggedness of small particle phase columns is also presented.

Thermo Scientific™ MAbPac™ SCX-10 Monoclonal Antibody Analysis Column

Thermo Fisher Scientific

MAbPac™ SCX-10 Columns for the High-Resolution Separation and Analysis of Monoclonal Antibody VariantsThe Thermo Scientific™ MAbPac™ SCX-10 column line has been expanded to include 3 and 5 μm particle sizes for fast, high-resolution analysis as well as rapid separation (RS) columns for bio-UHPLC separations. These smaller-particle-size columns provide the same high resolution as the 10 μm columns, but with significantly faster analysis time. In addition, salt or pH gradients can be used to provide fast high-resolution, high-throughput analysis of monoclonal antibody (MAb) variant samples. The MAbPac™ SCX-10 columns are strong cation-exchange columns designed specifically for the high-resolution, high-efficiency analysis of MAbs and associated variants. The unique nonporous pellicular resin provides exceptionally high resolving power, permitting the separation of MAb variants that differ by as little as one charged residue. Hydrophobic interactions with the resin are eliminated for very efficient peaks.MAbPac™ SCX-10 Monoclonal Antibody Analysis Column Features: Unequalled high-resolution separations Exceptionally high efficiency Highest resolution of monoclonal antibody variants Unmatched column-to-column and lot-to-lot reproducibility Hydrophobic interactions eliminated Ideal for stability testing Column Details: Chemistry: Sulfonic Particle Size: 3, 5, 10 μm Length: 50, 150, 250 mm ID: 2.0, 4.0, 4.6 mm

Thermo Scientific™ ProPac™ SCX and WCX Protein Column

Thermo Fisher Scientific

Strong and Weak Cation-Exchange Columns for High-Resolution Separation of Proteins and MAbsThe Thermo Scientific ProPac™ SCX column line has expanded to include the next generation ProPac SCX-20 column. This column provides unequalled high resolution of protein variants.ProPac SCX-10 and WCX-10 strong and weak cation-exchange columns are based on a nonporous core particle that results in high-efficiency peaks and high-resolution separations of protein variants. A hydrophilic layer prevents unwanted secondary interactions. A grafted cation-exchange surface provides pH-based selectivity control and fast mass transfer for high-efficiency separation and moderate capacity.ProPac® SCX and WCX Protein Column Features: Characterization and quality control assessment of monoclonal antibodies and other proteins Unequalled resolution High-resolution separations with excellent sample recovery Unmatched column-to-column and lot-to-lot reproducibility Useful for characterization of related protein variants (e.g., deamidation and MAb lysine truncation variants) Highly-efficient peaks with excellent sample recovery Available in different dimensions and formats for flexibility in method development Column Details: Length: 50, 100, 150, 250 mm ID: 2.0, 4.0, 9.0, 22 mm
