HepatoPac™: A Bioengineered Micro-Liver Platform for Predictive Drug Metabolism and Toxicity Studies

3 Nov 2015

Primary hepatocytes display a precipitous decline in phenotypic functions when cultured in a sandwich of extracellular matrix proteins (i.e. collagen, Matrigel). This severely limits the ability of these model systems to accurately predict clinical outcomes. This application note describes a human liver model with precise microscale cyto‐architecture and optimal stromal interactions (HepatoPac™) that displays high levels of liver‐specific functions for several weeks in vitro and has been shown to improve the predictions of clinical outcomes as compared to traditional approaches.

Cell / Tissue CultureCell culture or tissue culture is used to study the biology of cells or tissues and to isolate cellular products in an environment which can be manipulated and well defined. Accurately control your culture environment with bioreactors or culture incubators, bind your cells to a surface or together with an extracellular matrix. Distinguish cell types with differential media or proliferate cells with certain characteristics using selective media. Enrich your media with supplements such as growth factors, sera and vitamins. High-quality cells, optimized for your applications, alongside optimized cell substrates, growth medium and supplements, are critical for experimental success. Explore a range of cells suitable for your applications, including isogenic cell lines, competent cells, induced pluripotent stem cell (iPSC)-derived cell lines, fungal/bacterial/mammalian cell lines, stem cells and cancer cell lines. This can reveal information on cell health (apoptosis, cytotoxicity, viability and proliferation assays), cell metabolism, cell migration and cell signaling mechanisms. Find the best cell-based assay products, kits and equipment with our peer reviewed product directory: compare products, check customer reviews and receiving pricing direct from manufacturers. In VitroIn vitro refers to experiments conducted outside living organisms, often in controlled lab environments such as petri dishes or test tubes. In vitro models are widely used in drug testing, cell biology, and disease research. It involves the identification and quantification of cellular metabolites using analytical technologies such as GC, HPLC, NMR, and LC/MS.Drug TestingIt is essential to test the efficacy and toxicity of drugs at every stage of their development to ensure that a pharmaceutical product is fit for purpose and safe for use before reaching the stage of mass production and distribution.Protein InteractionsProtein interactions are essential for understanding cellular processes, as proteins work together to carry out biological functions. Studying protein interactions can provide insights into disease mechanisms, drug development, and cell signaling pathways. Explore the best tools for protein interaction research in our peer-reviewed product directory; compare products, check reviews, and get pricing directly from manufacturers.HepatocytesDrug MetabolismDrug metabolism is the enzymatic conversion within the body of pharmaceutical substances into other chemical compounds. The primary site for drug metabolism is the liver.
