Glycoprotein Monosaccharide Analysis Using HPAE-PAD with Eluent Generation
18 Mar 2015This study is intended as a guide for investigators interested in validating a high-performance anion-exchange chromatography with pulsed amperometric detection (HPAE-PAD)-based method for monosaccharide composition analysis. Described here are the configuration, typical performance, and precautions pertaining to use of an HPAE-PAD system for the monosaccharide analysis of glycoproteins. A Thermo Scientific Dionex ICS-5000 system with electrolytically generated eluent and a disposable gold on polytetrafluoroethylene (Au on PTFE) working electrode are used.
