Generation of mesenchymal stromal cell precursors in a 3D hollow fiber placenta co-culture model

16 Feb 2022

3D hollow fiber bioreactors provide a unique cell culture environment that closely recapitulates the in vivo microenvironment. Cells are in close proximity, secreted autocrine factors are concentrated, and cells do not require splitting so that cell-to-cell interactions are enhanced and allowed to develop over time.

In this white paper, FiberCell Systems demonstrates the generation of an in vitro placenta model using a hollow fiber bioreactor. This 3D co-culture of placenta-derived cells provides a potential source of mesenchymal stromal cell precursors and can be maintained for several months of continuous collection. Cell / Tissue CultureCell culture or tissue culture is used to study the biology of cells or tissues and to isolate cellular products in an environment which can be manipulated and well defined. Accurately control your culture environment with bioreactors or culture incubators, bind your cells to a surface or together with an extracellular matrix. Distinguish cell types with differential media or proliferate cells with certain characteristics using selective media. Enrich your media with supplements such as growth factors, sera and vitamins. High-quality cells, optimized for your applications, alongside optimized cell substrates, growth medium and supplements, are critical for experimental success. Explore a range of cells suitable for your applications, including isogenic cell lines, competent cells, induced pluripotent stem cell (iPSC)-derived cell lines, fungal/bacterial/mammalian cell lines, stem cells and cancer cell lines. Biological materials used include cells, enzymes and organisms. Usually bioprocessing requires a batch or continuous bioreactor such as a fermentor or cell culture system. The advantages of using a reactor include high productivity, easy configuration, adjustable values and automation.3D Cell CultureCulturing cells in three dimensions allows more physiologically relevant observations to be made regarding cellular interactions and gene expression. 3D models provide a more accurate representation of cells in vivo and are more stable than 2D cultures, making them very useful for long-term studies.FibersMesenchymal Stem Cells
