Extraction of Benzodiazepines From Human Urine Using ISOLUTE® SLE+ in Column Format Prior to GC-MS analysis

10 Jul 2012

This application note describes the extraction of a range of benzodiazepines from human urine using ISOLUTE SLE+ supported liquid extraction columns prior to derivatization and GC-MS analysis. Benzodiazepines are psychoactive drugs used to treat various conditions including anxiety, insomnia, seizures and alcohol withdrawal. This application note details an optimized extraction protocol for the use of ISOLUTE SLE+ supported liquid 1 mL sample volume columns to extract a range of benzodiazepines from human urine. This simplified and efficient extraction method has analyte recoveries from 90% with LOQs as low as 40 ng/mL. The application has also been performed with halogenated and non-halogenated extraction solvents in the interests of providing for green chemistry.

ISOLUTE SLE+ Supported Liquid Extraction Products

Biotage

ISOLUTE® SLE+ plates and columns enable efficient supported liquid extraction of analytes from whole blood, plasma, serum, urine, and oral fluid. The simple load-wait-elute method delivers 99.9% removal of proteins and phospholipids without offline steps, offering superior recovery, reproducibility, and cleaner extracts than liquid-liquid extraction.

LC-MSLC-MS (liquid chromatography-mass spectrometry) systems and equipment are used for separation and quantitative analysis of complex mixtures, combining liquid chromatography and mass spectrometry. Quantify proteins, contaminants, pesticides or screen for drug metabolites with a high level of sensitivity. LC-MS systems and equipment include reverse phase, normal phase and specialized columns integrated with various MS detectors such as time-of-flight (TOF), quadrupole, orbitrap or ion trap mass analyzers. LC-MS/MS instruments equipped with a qTOF or triple quadrupole analyzer give greater sensitivity and resolving power to your analysis. Multiple formats are available for conducting SPE, including prepacked SPE cartridges, disks and microplates, as well as SPE sorbent powders for manual packing. SPE systems can be used to automate the process and extract multiple samples at once. Solid-phase microextraction (SPME) and supported liquid extraction (SLE) products are also available in the same formats.
