The factors contributing to the potency of irreversible inhibition are poorly understood. This application note details Dr. Schwartz’s method for combining a reversible streptavidin-biotin capture method with the real-time direct observation of binding/reaction. It utilizes the Pioneer FE system from ForteBio and combines this with biochemical data to provide a full assessment of the inhibitor’s protein association, dissociation and reactivity. This ‘best of both worlds’ strategy provides the benefits of both biochemical and biophysical methodologies.
