Application of an Improved High-Throughput Lipid Extraction Method for the Analysis of Changes in Brain Lipid Species in Parkinson's Disease

Traditional lipid extraction techniques are the bottleneck for modern shotgun lipidomic studies. To overcome this problem, protocol comparisons were made between the well-established Folch extraction (using glass-glass homogenization) and a modified method that employed mechanical homogenization (Precellys-24, Bertin Technologies) and replaced chloroform with MTBE. This significantly reduced sample handling time and increased efficiency compared to glass-glass homogenizing.
