Analysis of Pesticide Residues, Mycotoxins and Potency in Cannabis using QuEChERS Extraction, ChloroFiltr® dSPE Cleanup and LC-MS/MS

22 Oct 2017

This application note outlines an all-in-one QuEChERS method for the simultaneous analysis of cannabis for 47 pesticides, 5 mycotoxins and 3 cannabinoids. Sample purification is carried out using UCT’s new cleanup product SpinFiltr™.

SpinFiltr

UCT, Inc.

The latest addition to UCT’s QuEChERS line puts a new “spin” on dSPE. SpinFiltr™ combines the practice of conventional dSPE with the added benefit of ultrafiltration. UCT’s new format of dSPE salts paired with a 0.2µm filtration device, simultaneously removes unwanted matrix compounds and filters samples without the need for any additional steps. With traditional dSPE, analysts must pay close attention when removing the purified extracts from tubes as to not disrupt the layers formed during centrifugation. SpinFiltr™ not only eliminates this problem, but also allows analysts to recover additional sample without the salts getting in the way!  Simply:  Add Vortex Spin Salts and unwanted matrix stay in the top chamber while purified extract is filtered and collected in the bottom portion of the tube. The chamber is discarded and analysts are left with only purified extract that is ready for analysis.

ChloroFiltr® for QuEChERS dSPE Clean-Up

UCT, Inc.

Chlorophyll in QuEChERS extracts must be removed to reduce potential instrument downtime. This is removed from extracts using graphitized carbon black (GCB) Unfortunately, GCB also removes planar analytes reducing analytical accuracy. ChloroFiltr®, used in place of GCB in dSPE tubes, removes unwanted chlorophyll without removing pesticides or environmental analytes. No change in procedure are required.

LC-MSLC-MS (liquid chromatography-mass spectrometry) systems and equipment are used for separation and quantitative analysis of complex mixtures, combining liquid chromatography and mass spectrometry. Quantify proteins, contaminants, pesticides or screen for drug metabolites with a high level of sensitivity. LC-MS systems and equipment include reverse phase, normal phase and specialized columns integrated with various MS detectors such as time-of-flight (TOF), quadrupole, orbitrap or ion trap mass analyzers. LC-MS/MS instruments equipped with a qTOF or triple quadrupole analyzer give greater sensitivity and resolving power to your analysis. It is most commonly used by analytical chemists studying pesticide residues in food.PurificationPurification techniques are essential for isolating compounds of interest from complex mixtures, such as proteins, nucleic acids, and chemicals. These techniques are critical in biochemistry, drug development, and environmental studies. They are classed as persistent organic pollutants (POPs) and can be harmful to humans. The Environmental Protection Agency (EPA) currently regulates the use of pesticides in the USA. MycotoxinsToxic chemicals such as Aflatoxin and Alkaloids that are produced by fungi are known as mycotoxins. In humid conditions, fungi can proliferate and colonize on crops to levels that can be damaging to human health. CannabinoidsCannabinoids are compounds found in cannabis that have therapeutic effects, such as pain relief, anti-inflammatory properties, and potential neuroprotective benefits. Research into cannabinoids is growing, particularly in the fields of pain management, neurological diseases, and cancer treatments.
