Analysis of Migration Using siRNA and the Oris™ Cell Migration Assay

21 Aug 2011

To study migration in cell lines siRNA and the ORIS™ TriCoated Cell Migration Assay were combined. This assay from Platypus offers certain advantages over traditional migration assays. U2OS cells were transfected with a control non-targeting siRNA or with three different siRNA pools targeting specific proteins. Fluorescence-based signal curves for tissue culture treated, Collagen I and Fibronectin coated wells were monitored with a POLARstar Omega microplate reader from BMG LABTECH.

RNA InterferenceRNA interference (RNAi) uses siRNA or miRNA for transcriptional silencing, gene knockdown and regulation of gene expression. RNAi requires chemical synthesis, introduction of DNA vectors into cells, an assay of RNAi effects and RNAi quantification or analysis. Consider target sequence selection, reagent preparation, controls, high specificity and effectiveness and low non-specific gene knockdown.
