An In Vitro Screening Tool for Predicting Clinical CYP3A4 Induction

2 Dec 2015

Induction‐mediated drug‐drug interactions need to be carefully characterized in vitro for drug candidates in order to determine and/or predict their induction potential in patients. Currently, both the FDA and EMA recommend using primary human hepatocytes as an in vitro test system for this purpose. In this poster, Corning HepatoCells (derived from primary human hepatocytes) were evaluated for its applicability as an in vitro screening tool for predicting clinical CYP3A4 inducers.

High-Throughput ScreeningHigh-throughput screening (HTS) is an automated drug discovery technique for identification of active compounds against a compound library. Use HTS readers and integrated assay preparation / analysis workstations to screen your compounds. Identify active compounds against various HTS libraries, including membranes, proteins and peptides and HTS cell lines. Find the best high-throughput screening products in our peer-reviewed product directory: compare products, check customer reviews and receive pricing direct from manufacturers.Cell-Based AssaysCell-based assays are used to monitor the presence, quantity and activities of a desired cellular analyte including drug molecules or biomarkers. This can reveal information on cell health (apoptosis, cytotoxicity, viability and proliferation assays), cell metabolism, cell migration and cell signaling mechanisms. Find the best cell-based assay products, kits and equipment with our peer reviewed product directory: compare products, check customer reviews and receiving pricing direct from manufacturers.Clinical ChemistryBiochemistry (or clinical chemistry) involves the analysis of bodily fluids using chemical tests. Techniques used include HPLC, chromatography, spectroscopy, mass spectrometry, immunochemical, electrophoresis, turbidometric / spectrophotometric assay, MRI and ISE analysis. Tests are often carried out on plasma or serum but urine (urinalysis) and fecal specimens are also processed.Clinical TrialsClinical Trials, an essential part of drug discovery process, assess the safety and effectiveness of a new medication or device in the pharmaceutical industry. Clinical Trials are a phased process (Phase 0, Phase I, Phase II, Phase III and Phase IV) which begins after initial preclinical testing. In VitroIn vitro refers to experiments conducted outside living organisms, often in controlled lab environments such as petri dishes or test tubes. In vitro models are widely used in drug testing, cell biology, and disease research. Explore in vitro research tools in our peer-reviewed product directory; compare products, check reviews, and get pricing directly from manufacturers.ScreeningUsing robotics, data processing and control software, liquid handling devices and sensitive detectors, screening allows a researcher to quickly conduct millions of chemical, genetic or pharmacological tests.Drug TestingIt is essential to test the efficacy and toxicity of drugs at every stage of their development to ensure that a pharmaceutical product is fit for purpose and safe for use before reaching the stage of mass production and distribution.HepatocytesHepatotoxicity
