A New Quantitative Microplate-Based Multiplex Technology

In this application note from R&D Systems, a method is described to quantify several different proteins in an experimental sample. The Mosaic ELISA Human Cytokine Panel 1 is used to detect multiple analytes simultaneously in PHA-treated human peripheral blood mononuclear cells (PBMCs). By combining the design and specificity of the traditional ELISA with multiplex technology, the Mosaic ELISA Human Cytokine Panel 1 and Human Growth Factor Panel 1 are designed to measure the levels of up to 8 different cytokines or growth factors simultaneously in a single sample of cell culture supernate, serum, or plasma.

The Mosaic ELISA Human Growth Factor Panel 1 is an excellent tool for the detection of 6 different growth factors in the same sample. Mosaic ELISA kits employ multiplex microarray technology to provide an accurate, efficient, and economical alternative to conducting multiple traditional ELISA experiments. Growth factor is a term used to describe a broad range of structurally diverse molecular families and individual proteins best known for their ability to enhance cell proliferation and growth. They have an array of putative functions during development including regulating tissue morphogenesis, angiogenesis, cell differentiation, and neurite outgrowth. They also play important roles in the maintenance of tissue homeostasis and wound healing in the adult. Their activities are mediated via binding to transmembrane receptors that often contain cytoplasmic tyrosine kinase domains. Cytokines are pleiotropic extracellular signaling molecules that influence myriad physiological and pathological processes. Released from a variety of cells, their actions affect cellular growth, differentiation, gene expression, migration, immunity, and inflammation. In most biological processes, multiple cytokines operate in a large network where the action of one cytokine is regulated by the presence or absence of other cytokines. Studying the underlying signaling cascades is further complicated by the fact that many cytokines modulate the production of other cytokines.
