Application Area:

Antibody analytics

The possibility to use temperatures up to 90°C simplifies the development of analytical methods. Furthermore, a good peak shape can be obtained without the addition of TFA, which means that I have fewer problems when using it for mass spectrometry. I also like the good customer support in the form of competent advice, quick answers and the uncomplicated solutions for special cases.

Application Area:

HPLC

This HPLC column gives better separation and peak shape for many components. We found some isomers also got separated. We are very happy with the product and the supplier gave us very good support by giving us a demo-column. We were able to determine the column's efficiency and know whether the column could separate the impurities in our product. Thanks to YMC, most of our HPLC methods are developed with this product.

Robust and reproducible UHPLC/HPLC columns with a wide range of stability towards pH (1-12), temperature and 100% aqueous eluents. YMC-Triart columns offer solutions for every analyte covering the range from very polar to very hydrophobic substances as well as acidic, neutral and polar compounds. Full scalability is ensured due to particle sizes from 1.9 to 20 µm. Separation of hydrophobic and medium polar to very polar analytes.

YMC-Triart and YMC-Triart Bio are robust and reproducible UHPLC/HPLC columns for BioLC which are highly stable towards pH and temperature (up to 90 °C). These columns with different pore sizes (12 or 30 nm/120 or 300 Å) are recommended for analysis of proteins, antibodies, peptides and oligonucleotides. Wide-pore (300 Å) YMC-Triart Bio phases are specially designed for large biomolecules, while the classic YMC-Triart C18 or C8 phases are suitable for peptides and oligonucleotides. Using YMC-Triart Bio C18 or C4 columns with formic acid eluents allows optimal peak shapes for antibodies and proteins to be obtained, which make these phases equally highly suitable for LC-MS as the classic YMC-Triart phases.

YMC-Triart & YMC-Triart Bio Columns Features:

  • An ideal choice for proteins, antibodies, peptides, and oligonucleotides
  • Superior lot-to-lot reproducibility
  • High matrix tolerance due to the increased surface, e.g. plasma
  • Excellent choice for LC-MS
  • C18 & Bio C18 compatible with 100% aqueous eluents

YMC-Triart & YMC-Triart Bio Columns Characteristics:

  • YMC-Triart C18 and C8: 12 nm/120 Å pore size
  • YMC-Triart Bio C18 and Bio C4: 30 nm/ 300 Å pore size
  • Particle Sizes: 1.9, 3, 5µm
  • pH range: 1-12 (C18 or C8)
  • Max. Temp: up to 90°C
  • Bioinert hardware available

DAR determination using third generation conjugation method

Antibody-drug conjugates (ADCs) represent a rapidly evolving class of targeted therapies, with three distinct generations of conjugation methods. The first generation utilises lysine residues for drug attachment, while the second generation relies on cysteine coupling. The third generation, marks a significant advancement with site-specific conjugation, where cytotoxic drugs are attached at defined positions on the antibody, leading to a more homogeneous ADC with a precise drug-to-antibody ratio (DAR) of either 2 or 4. YMC Europe demonstrates the accurate DAR determination of a site-specific ADC using a YMC-Triart Bio C4 column, a robust wide-pore hybrid silica-based column designed to deliver reliable, high-resolution separations.

YMC biochromatography columns

In this application note, YMC Europe provides a comprehensive overview of its bio-chromatography columns, including best-use scenarios for reversed-phase (RP) size exclusion (SEC), ion exchange (IEX), and hydrophobic interaction (HIC) columns. Additionally, YMC has included a guide on phase selection for proteins, monoclonal antibodies, and oligonucleotides.

How to influence oligonucleotide analysis by IP-RP

In this application note, YMC presents experimental data on oligonucleotide analysis using the YMC-Accura Triart Bio C18 column. The purity and quality of oligonucleotides needs to be controlled closely since impurities can affect their efficacy. The analysis of oligonucleotides is highly challenging due to their negatively charged phosphate groups in the backbone. Ion pair reversed phase liquid chromatography (IP-RP) is considered the gold standard for oligonucleotide analysis.

Generic Analyses of Different Monoclonal Antibodies with RP-(U)HPLC

Monoclonal antibodies (MAbs) are immunologically active proteins, which bind specifically to certain cells or proteins, this stimulates the immune system to attack those targets. MAbs are very important for the treatment of different kinds of cancer and autoimmune diseases. Nowadays, a broad variety of therapeutically antibodies are available on the market and several more are in research and development. In this application note, shows how to achieve robust chromatographic results for commercially available MAbs.
