Thermo Fisher Scientific Inc., the world leader in serving science, today announced that it will feature its Thermo Scientific Dionex UltiMate 3000 Biocompatible Rapid Separation (BioRS) liquid chromatography system during Pittcon 2013. This system is designed to combine the performance advantages of ultrahigh-performance liquid chromatography (UHPLC) with the corrosion resistant flow path demanded by the analysis of proteins, peptides, nucleic acids and biotherapeutics. Thermo Fisher will showcase the new system within booth 1835 during Pittcon 2013, being held in Philadelphia from March 17-21, 2013.

“We’ve designed this system to accelerate research into promising biotherapies, such as monoclonal antibody variants, and also to support biopharmaceutical research and QA/QC, including glycan analysis,” said Wulff Niedner, director, HPLC product marketing, for Thermo Fisher Scientific. “There is strong demand for a true high-performance biological separation platform, offering capabilities beyond reverse phase UHPLC. This combination of state-of-the-art technology and low-dispersion flow path is engineered to provide the high-peak capacity and sensitivity needed to analyze complex biomolecules.”

The system was developed for reliability and durability, even with harsh solvents used for many bioseparations. The flow path is built of corrosion-resistant materials, enhancing robustness and extending the life of biocolumns, even under high salt or extreme pH conditions. In addition, the materials used are designed to significantly reduce the risk of analytes interacting with internal surfaces of the flow path, protecting data quality.

The UltiMate 3000 BioRS system is powered by UltiMate 3000 Rapid Separation technology to support the high pressures required to separate many bioanalytes on high resolution UHPLC columns. The system is engineered to operate at pressures as high as 1,000 bar (15,000 psi) and flow rates up to 8 mL/min. This wide flow/pressure footprint supports many biomolecule characterizations, including development, identification and quality control. The tool-free biocompatible Thermo Scientific Dionex Viper fingertight fitting system is designed to facilitate connections without dead volume to combat peak-broadening effects.

The modular system offers many configuration options to accommodate most large molecule workflows. These include binary, quaternary and dual gradient mobile phase delivery, as well as parallel and tandem LC configurations.

Columns designed to optimize system performance
Thermo Fisher Scientific also chose Pittcon 2013 to introduce columns developed specifically to complement the performance of the UltiMate 3000 BioRS system. The Thermo Scientific MAbPac SCX-10 RS column is designed for UHPLC separations of monoclonal antibodies. The Thermo Scientific DNAPac PA200 RS column can be used to analyze single nucleic acids and isomers to yield important structural information.

Both feature PEEK linings, providing highly inert flow paths designed to remain stable at UHPLC pressures, and ultra-stable packed beds designed for excellent retention reproducibility and long life.
Another biocolumn choice is the new Thermo Scientific Dionex GlycanPac AXH-1 column, designed to enhance glycan analysis with simultaneous charge, size and polarity HPLC separations.

UHPLC and HPLCHigh performance liquid chromatography (HPLC) and ultra high performance liquid chromatography (UHPLC), also known as UPLC, are analytical techniques used to separate, identify and quantitate components of complex mixtures including biological samples such as proteins and lipids as well as chemical mixtures of pesticides, drugs and oils. Both techniques are liquid chromatographic methods but differ by operating pressures (HPLC < 6000 psi < UHPLC ). Components of HPLC and UHPLC systems include columns, detectors, pumps, autosamplers and column heaters.
