Denator AB announced today the release of a new kit to ensure scientists obtain a consistent, representative picture of protein profiles when analyzing heat-stabilized tissue using standard two-dimensional gel electrophoresis (2D-GE). The new Stabilizor™ 2D-GE Extraction Kit when used together with the company’s Stabilizor™ system enables users to heat-stabilize, homogenize and extract proteins from tissue samples following a validated protocol developed to maximize protein recovery.

The Stabilizor™ system, based on Denator’s heat-stabilization technology, has been shown to stop biological changes in tissue from the moment of sampling, this prevents the enzymatic processes that alter protein states and cause protein degradation. Earlier this year the company introduced the Stabilizor™ Peptide Extraction Kit to prepare and extract peptides from heat-stabilized tissue prior to LC-MS analysis.

Olof Sköld, CEO at Denator, explained further: “Our goal is to ensure that Stabilizor™ system users can benefit fully from the advantages of using heat stabilization upstream and have full confidence in the quality of data from analysis downstream. The new kit offers the convenience of having all necessary accessories and solutions ready to use, and, most importantly, includes a validated protocol designed to ensure optimal extraction of proteins from heat-stabilized samples prior to analysis. We are very pleased to offer kits that support two of the most common analytical techniques, in use today - 2D-GE and LC-MS.”

Implementation of the Stabilizor™ solution in laboratory workflows is reflected in a growing number of peer-reviewed publications particularly from proteomic laboratories. Rapid heat stabilization enables scientists to reveal true protein profiles, detect endogenous peptides and preserve phosphorylation states.

ElectrophoresisElectrophoresis equipment is used to separate mixtures of protein, DNA or RNA, based on their electric charge, size and other physical characteristics, by passing them through a medium such as a polyacrylamide gel, an agarose gel or a capillary tube. Electrophoresis equipment includes horizontal and vertical gel electrophoresis chambers, isoelectric focusing systems, 2D electrophoresis and capillary electrophoresis instruments. Precast gels with a gradient can be used or gels can be hand cast. Samples are run alongside ladders or markers to identify the approximate size of a molecule. Separated proteins and nucleic acids in the gel can be stained or probed with fluorescent markers and then imaged and detected using gel documentation instruments, transilluminators, densitometers and scanners. Consequently, the importance of high-quality DNA/RNA extraction and purification equipment cannot be underestimated. Many purification kits are available and are typically optimized for nucleic acid type and source, including plasmid DNA, genomic DNA, mRNA, RNA and viral nucleic acid purification kits. Automated extraction and purification of nucleic acids can be implemented with magnetic bead separator instruments or high-throughput purification workstations.
