TaKaRa DNA Kination Kit是对非磷酸化DNA片段(单链及双链),如:PCR产物、Linker、Adaptor、Primer等的5'末端进行磷酸化的试剂盒。试剂盒中含有T4 Polynucleotide Kinase和ATP,与TaKaRa公司独自开发的5'末端高效磷酸化反应液相结合,可以对DNA片段(单链及双链)的5'末端进行高效磷酸化。
试剂盒中的pSIMPLE-18 EcoR V/BAP Vector是TaKaRa公司根据pUC 18载体改建而成的平滑末端去磷酸化载体,本载体消去了pUC 18载体上的多克隆酶切位点,但仍保持着β-半乳糖苷酶的正常表达,进行DNA克隆时仍可以利用α-互补性进行蓝白菌落筛选。由于消去了多克隆酶切位点,克隆后的DNA片段将无法用载体上的酶切位点切下,此时需要在DNA片段的两端导入合适的酶切位点,使用导入的DNA片段上的酶切位点进行DNA切断时,酶切反应将不会受到原有载体上其它多克隆酶切位点上的限制酶影响,可以大大提高酶切效率,增加亚克隆的成功率。
试剂盒中还含有Control Insert 3以及高效连接液Ligation Mix,可以进行Control实验。而高效连接液Ligation Mix可以在极短时间内(5分钟左右)完成连接反应,且其连接液可以直接用于细菌转化,大大方便了克隆实验。
