AnaSpec Announces SensoLyte® Deubiquitination Assay Kits

Product news

AnaSpec, EGT Group, is pleased to introduce the SensoLyte® 440 and 520 Deubiquitination Assay Kits. The former kit provides a convenient assay for inhibitors/activators screening in deubiquitinylation or for continuous assay of enzyme activity. The latter kit, employs a peptide substrate conjugated to a green fluorophore, and is ideal for inhibitor/activator screening in deubiquitinylation or for continuous assay of enzyme activity.

A natural substrate, ubiquitin, coupled with the AMC (7-amino-4-methylcoumarin) fluorophore is used in the SensoLyte® 440 kit. Upon cleavage by a deubiquitin protease, this substrate generates a blue fluorophore that can be detected at excitation/emission=354/442 nm. The substrate supplied in the kit is exquisitely sensitive for ubiquitin hydrolases (UCHs). The detection limit can reach as low as 3.7 pg/mL.

Within the SensoLyte® 520 Deubiquitination Assay Kit from AnaSpec, bright green fluorescence is generated upon protease cleavage and can be detected at excitation/emission=490/520 nm. The long wavelength spectra of the green fluorophore released provides less interference from other reaction components.

Assay AssemblyAssay Assembly is technique used in drug discovery to develop assays to test the cytotoxicity, genotoxicity, or other activities of a compound on a cell. Assay assembly requires chip assembly, a delivery system and a detection and analysis method. Beneficial features of assay kits or automated systems include high-throughput, high speed and sensitivity and low signal to noise ratio.Protein PurificationProtein purification is a vital step in drug discovery, therapeutics, biotech and life science research. The purification process typically involves subcellular or membrane protein extraction with cell lysis kits, separation of proteins from cell debris by filtration or spin columns, and the isolation of proteins of interest from other proteins and impurities with affinity purification (including fusion protein tags and antibody binding proteins A, G and L), immunoprecipitation or chromatographic methods, such as ion exchange, size exclusion and immobilized metal affinity chromatography. All purification methods come in multiple formats for your laboratory needs, including agarose or magnetic beads, resins, columns and filter plates.
