Adeno-Associated Virus (AAV) vectors have developed to be one of the most commonly used vectors for gene transfer, representing a promising therapeutic approach to target inherited, genetic diseases, and many others. The gene therapy community, attempting to find treatments for a variety of genetic disorders faces several obstacles such as meeting the requirements from regulatory authorities as well as the time pressure caused by the rapidly growing market. Though the use of orthogonal methods is recommended for comprehensive characterization, rapid testing of individual and crude AAV samples during the development and manufacturing process is often difficult. Most precise analytical AAV tools are not convenient for single sample measurements and/or require highly purified samples.

PROGEN´s mission is to provide high-quality products that help advance new therapies and develop existing research processes safely, quickly, and affordably for the gene therapy community. For this reason, the PROGEN team developed the Dip´n´Check AAV kits, based on PROGEN’s expertise in AAV assay development and the urgent need for fast analytical methods in the growing field of AAV-based gene therapy.

The Dip´n´Check AAV lateral flow tests allow fast in-process control at every step of your process and can be used for the analysis of crude lysates as well as purified AAV samples. The kits have been developed by PROGEN based on its unique AAV particle antibodies which detect fully assembled AAV particles and allow the estimate of the actual AAV titer in your sample. These antibodies have widely been proven to be a suitable and reliable tool for the detection of intact AAV capsids.

Possible applications of the Dip’n’Check lateral flow tests include:

Comparison of AAV titers of different AAV preparations to e.g.

  • Select the most promising sample for further processing
  • Estimation of AAV titers in crude and purified AAV preparations to e.g.

  • Control efficiency of your processes
  • Pre-test AAV concentrations to hit the dynamic range of downstream analytics
  • Testing the binding of common AAV antibodies to an AAV capsid variant to e.g.

  • Validate suitability of antibody-based methods for further analysis
  • The rapid, semi-quantitative analysis of the Dip´n´Check AAV kits facilitate the acceleration of decision processes and help save precious time and sample in less than 20 minutes.

Just follow the simple 3-step process:

Prepare your sample by adding the included antibody solutions

    Incubate for 10 minutes

Add the lateral flow test stripe and get your result in just 10 minutes

Increase the probability for success and establish how to proceed best in the process early on.

    Key features:

    • Results in less than 20 minutes
    • Suitable for analyzing crude lysate and purified AAV samples
    • Ideal in-process control for purification processes for example
    • Pre-testing AAV concentrations for downstream analytics
    • Available for AAV1, AAV2, AAV3, AAV5, AAV6, AAV8, and AAV9

ELISAEnzyme-linked immunosorbent assays (ELISA), also known as enzyme immunoassays (EIA), are used for the detection and quantification of proteins, peptides and antibodies in a sample. ELISA often comes in kit format, with pre-selected antibody pairs to detect specific proteins or biomarkers of diseases such as diabetes and obesity, or cardiovascular and neurological disorders. Detection is made possible with chemiluminescent, fluorescent or colorimetric substrates and detection instruments such as microplate / ELISA readers. Other kits available include: Enzyme-linked immunospot (ELISPOT) assays for the quantification of protein-producing cells and chemiluminescent immunoassays (CLIA) kits.
